The major problems with these therapies is that the effects of the drug are not limited to the target tissue and lead to undesirable side effects. These side effects are really complex and are due to interactions of estrogen, its binding partner, estrogen receptor (ER,) and a whole network of other signaling proteins. Changes in these complex interactions are also the most likely cause of antiestrogen resistance in the breast cancer, when the standard drugs for ER-positive breast cancer are no longer effective. \u00a0Our lab wants to know why these drugs have different responses in different tissues and how this leads to adverse side effects and anti estrogen-resistant breast cancer. Answering these questions could lead us to new drugs with better profiles for breast cancer and hormone replacement therapies.
